メチル化DNA IP（MeDIP）は、hMeDIPの場合、5-メチルシトシン（5-mC）または5-ヒドロキシメチルシトシン（5-hmC）に対する抗体を使用したメチル化DNAのアフィニティ精製に基づいています。
メチルDNA IPの手法は、培養細胞または組織からのゲノムDNAが調製され、断片化された後に変性されます。 次に、5つのメチルシトシンに対する抗体と抗体結合ビーズを使用して、免疫選択と免疫沈降を行ないます。 単離および精製が行われた後、IP化されたメチル化DNAは、qPCR、増幅、マイクロアレイでのハイブリダイゼーション、または次世代シーケンシングなど、その後の解析に使用できます。
