Figure 1. Immunoprecipitation using the Diagenode monoclonal antibody directed against PABPC1
Immunoprecipitation was performed on total RNA isolated from 15 million 293T cells using 15 µg of the Diagenode monoclonal antibody against PABPC1 (Cat. No. C15200240) or with an equal amount of mouse IgG, used as a negative control. The immunoprecipitated RNA was subsequently analysed on a Bioanalyzer. Figure 1 shows the Bioanalyzer profile obtained with the negative control (upper left) and the PABPC1 antibody (upper right). The lower figure shows the gel image for the PABPC1 antibody, the negative IgG control, and the input (lane 1, 2 and 3 respectively). The marker (in bp) is shown on the left, the position of the 28s and 18s ribosomal RNA is indicated on the right.

Figure 2. Western blot analysis using the Diagenode monoclonal antibody directed against PABPC1
Whole cell extracts from Jurkat, HeLa, 293T, HL-60, A431 and CHO cells (lanes 1 to 6, respectively) were analysed by Western blot using the Diagenode monoclonal antibody against PABPC1 (Cat. No. C15200240) diluted 1:1,000 in PBS containing 1% skimmed milk. The position of the protein of interest is indicated on the right; the marker (in kDa) is shown on the left.

Figure 3. Immunoprecipitation using the Diagenode monoclonal antibody directed against PABPC1
Immunoprecipitation was performed on whole cell extracts from 293T cells using 2 µg of the Diagenode monoclonal antibody against PABPC1 (Cat. No. C15200240, lane 2). An equal amount of mouse IgG was used as a negative control (lane 1). The immunoprecipitated PABPC1 protein was subsequently detected by western blot with the PABPC1 antibody as described above.

    IF

Figure 4. Immunofluorescence using the Diagenode monoclonal antibody directed against PABPC1
HeLa cells were stained with the Diagenode monoclonal antibody against PABPC1 (cat. No. C15200240). Cells were fixed with 4% formaldehyde for 10 min at RT, permeabilized with 0.1% Triton X-100 for 10 min at RT and blocked with PBS containing 10% goat serum. The cells were immunofluorescently labeled with the PABPC1 antibody diluted 1:200 in blocking solution followed by a goat anti-mouse antibody conjugated to Alexa488 (green). The nuclei were counterstained with DAPI (blue).
