Nucelolin EMSA Kit

Key features: Nucleolin, isolated from HeLa cells, harbors an activity to efficiently interact with RNA in vitro, without changing the overall integrity of substrate RNA. In a standard reaction (10 ul), nucleolin (50 ng) forms complexes with virtually all RNA substrates (1 nM) provided. However, the optimal concentration of nucleolin should be emperically determined for each RNA substrate of researcher's interest. The following reagents will be provided:

• Nucleolin: 5 ug (0.1 mg/ml).
• 10x Reaction buffer (0.25 ml): 0.2 M Hepes-NaOH, pH 7.4, 1 M NaCl, 10 mM DTT, and 2 mg/ml BSA.
• 5x Loading buffer (0.5 ml); 0.1 M Tris-HCl, pH 7.4, 5 mM EDTA, 0.05% NP-40, 0.01% bromophenol blue, 0.01% xylene cyanol, and 50% glycerol
• Protein dilution buffer (0.5 ml): 20 mM Hepes-NaOH, pH 7.4, 50 mM NaCl, 0.1 mM EDTA, 0.5 mM PMSF, 0.2 mg/ml bovine serum albumin, 0.05% Noniodit P-40, a cocktail of protease inhibitors, and 20% glycerol.
