Key features: The use of an internal control DNA makes it possible to monitor the efficiency of in vitro chromatin assembly with target DNA of either linear or circular conformation. In vitro chromatin assembly kit contains the following reagents:

• 10x Reaction Buffer (200 ul): 0.2 M Hepes-NaOH, pH 7.4, 0.1 mM EDTA, 0.5 mM PMSF, and 10 mM DTT.
• Control DNA template (pNAS, 2 ug, 0.1 mg/ml)
• PCR primer mix (40 ul, 100 pmol/ul)
• 5 M NaCl (200 ul)
• HeLa core histones (50 ul, 2 mg/ml)
• Dilution Buffer (0.5 ml): 20 mM Hepes-NaOH, pH 7.4, 0.1 mM EDTA, 0.5 mM PMSF, and 1 mM DTT
• Storage Buffer (1 ml): 40 mM Hepes-NaOH, pH 7.4, 0.2 mM EDTA, 0.5 mM PMSF, 2 mM DTT, 0.1% NP-40, 0.6 mg/ml BSA, and 30% glycerol.
