Auto hMeDIP kit
Description
This kit has been designed to perform Hydroxymethylated DNA Immunoprecipitation using an antibody against 5-hydroxymethylcytosine (5-hmC) on our Automated Systems. It includes control sequences (5-hmC, 5-mC and unmodified cytosine DNA standards) and control primer pairs (internal and external controls), to assess the efficiency of your immunoprecipitation.

> Complete magnetic bead based kit
> Generate highly consistent results with internal controls in 24h
> Allows direct correlation between IP’d material & hydroxylmethylation status
> Including internal & external controls for QC of each reaction
> Mouse primers against Sf1 (hydroxymethylated gene in mouse)
> 5-hmC, 5-mC and unmethylated DNA
> Validated on Diagenode's Automated Systems

Figure. 1 µg of DNA from mouse ES cells was prepared and sonicated with the Bioruptor® to obtain DNA fragments of 300-500 bp. Unmethylated, methylated and hydroxymethylated spike DNA controls (included in the Auto hMeDIP kit) were used in the IP reaction together with the genomic sheared DNA samples. Enrichments were assessed by qPCR using specific primer pairs for the unmethylated, methylated and hydroxymethylated DNA sequences. Sfi1 is a gene that has been identified as being hydroxymethylated using hMeDIP-seq. Human DNA from U2OS was used as negative control DNA.
